The Agar Method: Preparation Continued

  1. Place the cone shaped tip piece in an appropriately labeled cassette. The cassette is then placed in fixative and processed as tissue.

     

  2. At embedding, slice the cellular button in half to form two semi-circles (see Figure 12).

     

  3. The two semi-circles must be embedded with their common cut side down, so that a section is cut through all layers of the centrifuged button. This is a critical step in the process. Improper embedding will nullify all the previous efforts.
