Cellular Material Entrapped in Mucus: Preparation

The following method can be used to prepare a specimen from material trapped in mucus:

  1. After all of the slides have been made, mix up the remaining portion of the sediment by tapping the container gently on the countertop.


  2. Add approximately 25 - 30 ml of 95% ethyl or reagent alcohol to the centrifuge tube and mix by inverting several times. Alternately, 10% neutral buffered formalin may be used, but it is a slower fixative than alcohol.


  3. Allow this to sit until the specimen is completely fixed (usually 15 - 30 minutes).
